ATHENS, Greece (AP) ? Coast guard officials say they have found the bodies of 18 people who were trying to cross illegally into Greece when their boat capsized early Friday.

A 20-year-old man found alive Friday told authorities there were 28 people on the crowded boat, including its Turkish owner, when it capsized off the northeastern island of Lesvos, near the Turkish coast. The man, who has been hospitalized, is the sole survivor found so far.

A coast guard spokeswoman said the migrants were "of Asian origin" without specifying further.

Coast guard vessels have been scouring the nearly calm seas east of Lesvos.

Tens of thousands of undocumented migrants cross into Greece each year, most through land and sea coming from Turkey.

Factories flex muscles post storm, inflation subdued

WASHINGTON (Reuters) - Factory output posted its sharpest increase in nearly a year in November as auto production staged a rebound, while consumer prices slipped, offering cautious optimism for the struggling economic recovery.

Factories have bounced back after being held down by Superstorm Sandy, which struck the East Coast in late October.

Despite last month's rise, factory production remained below highs reached earlier this year. Analysts said this subdued recovery and tame price pressures provide ample scope for the Federal Reserve to stay on its ultra-easy monetary policy path.

"This is an economy that still has a lot of slack and upside potential," said Robert Dye, chief economist at Comerica in Dallas. "There is a lot of dry tinder out there, the Fed has added to that with monetary policy and we have to get past the fiscal cliff issues to see if the dry tinder catches fire."

The "fiscal cliff" refers to the $600 billion in deep government spending cuts and tax hikes that will hit the economy next year if the Obama administration and Congress fail to agree on a less drastic plan to reduce budget deficits.

Manufacturing output rose 1.1 percent in November, the biggest gain since December 2011 and a rebound from a 1.0 percent drop in the prior month, the Federal Reserve said. It said production was lifted by a surge in motor vehicle output.

Hurricane Sandy had weighed on overall industrial output in October, but the rebound in November was stronger than economists had expected. Output at the nation's factories, mines and utilities taken together also jumped 1.1 percent after slumping 0.7 percent in October. It was the biggest gain in almost two years.

Separately, financial information firm Markit said its preliminary gauge of U.S. factory activity in December rose to 54.2, its highest level since April, from 52.8 in November.

"Going forward, output will largely be determined by what type of resolution is reached on the fiscal cliff," said Brett Ryan, a U.S. economist at Deutsche Bank Securities in New York.

"If policymakers do not come up with a solution, the recovery in industrial production may prove to be fleeting."

BENIGN INFLATION ENVIRONMENT

In a third report, the U.S. Labor Department said its consumer price index dropped 0.3 percent in November, the first decline in six months, as gasoline prices fell sharply.

The so-called core CPI, which excludes food and energy prices, edged up 0.1 percent after rising 0.2 percent in October. Although food prices rose 0.2 percent in a lagged response to the summer drought, price pressures remained tame.

"The inflationary backdrop remains very benign, providing the Fed with considerable breathing room to keep monetary policy accommodative," said Millan Mulraine, a senior economist at TD Securities in New York.

The central bank said on Wednesday it expected to hold interest rates near zero until the unemployment rate falls to at least 6.5 percent as long as inflation does not threaten to break above 2.5 percent.

U.S. stock prices ended lower on Friday as the data failed to dispel worry on Wall Street over the fiscal cliff. Prices for U.S. government debt rose as traders saw the inflation data supporting an easy Fed policy. The dollar fell from a near nine-month high against the yen and dropped for a fifth straight day against the euro.

GASOLINE COSTS DROP

In the 12 months through November, consumer prices increased 1.8 percent, the smallest gain since August and a slowdown from the 2.2 percent rise in the period through October. It has slowed from nearly 3 percent in January.

Last month, gasoline prices tumbled 7.4 percent, the largest drop since December 2008, after falling 0.6 percent in October.

The drop in gasoline prices eased some of the strain on household budgets. The Labor Department said inflation-adjusted average weekly earnings rose 0.5 percent last month, reversing October's 0.5 percent fall.

Away from gasoline, the cost of apparel fell for the first time in two months, while new motor vehicle prices saw their first rise since August. Auto prices could have been lifted by a spike in demand as people replace vehicles destroyed by Sandy.

Prices for used cars and trucks fell for a fifth straight month, while housing costs edged up.

In the 12 months through November, the core CPI increased 1.9 percent after rising 2.0 percent in October.

"The current disinflationary trend, and the fact that wage and upstream price pressures are almost non-existent, suggests that core inflation will probably remain below the Fed's target for most of 2013," said Jeremy Lawson, a senior economist at BNP Paribas in New York.

The Fed targets an inflation rate of 2.0 percent, but uses a separate gauge that tends to run a little lower than the CPI.

Experimental agent briefly eases depression rapidly in test

A drug that works through the same brain mechanism as the fast-acting antidepressant ketamine briefly improved treatment-resistant patients' depression symptoms in minutes, with minimal untoward side effects, in a clinical trial conducted by the National Institutes of Health. The experimental agent, called AZD6765, acts through the brain's glutamate chemical messenger system.

Existing antidepressants available through prescription, which work through the brain's serotonin system, take a few weeks to work, imperiling severely depressed patients, who can be at high risk for suicide. Ketamine also works in hours, but its usefulness is limited by its potential for dissociative side-effects, including hallucinations. It is being studied mostly for clues to how it works.

"Our findings serve as a proof of concept that we can tap into an important component of the glutamate pathway to develop a new generation of safe, rapid-acting practical treatments for depression," said Carlos Zarate, M.D., of the NIH's National Institute of Mental Health, which conducted the research.

Zarate, and colleagues, reported on their results online Dec. 1, 2012 in the journal Biological Psychiatry.

AZD6765, like ketamine, works by blocking glutamate binding to a protein on the surface of neurons, called the NMDA receptor. It is a less powerful blocker of the NMDA receptor, which may be a reason why it is better tolerated than ketamine.

About 32 percent of 22 treatment-resistant depressed patients infused with ASD6765 showed a clinically meaningful antidepressant response at 80 minutes after infusion that lasted for about half an hour ? with residual antidepressant effects lasting two days for some. By contrast, 52 percent of patients receiving ketamine show a comparable response, with effects still detectable at seven days. So a single infusion of ketamine produces more robust and sustained improvement, but most patients continue to experience some symptoms with both drugs.

However, depression rating scores were significantly better among patients who received AZD6765 than in those who received placebos. The researchers deemed this noteworthy, since, on average, these patients had failed to improve in seven past antidepressant trials, and nearly half failed to respond to electroconvulsive therapy (ECT).

The patients reported only minor side effects, such as dizziness and nausea, which were not significantly different from those experienced with the placebo.

Zarate and colleagues say their results warrant further trials with AZD6765, testing whether repeated infusions a few times per week or higher doses might produce longer-lasting results.

Uncovering a flaw in drug testing for chronic anxiety disorder

Dec. 13, 2012 ? Pre-clinical trials -- the stage at which medications or therapies are tested on animals like laboratory mice -- is a crucial part of drug development. It's only then that scientists can assess benefits and side effects before a drug is administered to patients.

Now, Prof. Ilan Golani of Tel Aviv University's Department of Zoology and Sagol School for Neuroscience and his fellow researchers Prof. Yoav Benjamini of TAU's Department of Statistics and Operations Research and the Sagol School of Neuroscience, and Dr. Ehud Fonio of the Weizmann Institute are questioning the animal models used for measuring chronic disorders such as Generalized Anxiety Disorder. Using a hundred-times longer experimental time-frame and comparing lab mice with wild mice, the researchers found that the lab mice used do not actually experience chronic anxiety.

Instead, the researchers found that the behavior exhibited by the mice in the first minutes of an experiment is only a temporary response to a new environment. With time, the mice revert back to their true temperaments -- lab mice have calm temperaments and wild mice are anxious. This discovery, which has been reported in the journals PLoS One and Nature Methods, could explain why most candidate drugs developed using this mouse model have poor therapeutic value in treating human brain disorders, says Prof. Golani.

Back to the wild

Because of their genetic similarity to humans, mice are the most commonly used lab animals. Countless hours and billions of dollars have been spent developing mouse models, whose genes can be engineered to mimic human diseases and disorders. And while many of these models have made invaluable contributions to the advancement of research into Parkinson's disease and various cancers, others have proven less effective.

A chronically anxious mouse model is crucial to test anti-anxiety therapies, says Prof. Golani. Currently, scientists use a specific strain of lab mice thought to be particularly anxious. During experiments, the mice are placed in a novel environment and monitored for signs of anxiety such as staying in sheltered rather than exposed space, for example. After drugs are administered, the mice are observed for a reduction in anxious behaviors.

The researchers compared the lab mouse strain now used for Generalized Anxiety Disorder testing to a strain of first-generation wild mice born in captivity. The time-frame of the experiment was extended from a few minutes to as long as 45 hours. Though the lab mice appeared more anxious at first, the scientists discovered that the lab mice eventually settle into calm behavior, while the wild mice exhibit consistently more anxious behavior: "In nature, mice must always be on high alert or they will get preyed upon," explains Prof. Golani.

They believe that the current Generalized Anxiety Disorder methodology for animal models should be revised because of three main experimental fallacies: use of the wrong animal, too short a time-frame, and analysis made at the wrong points in the experiment. These prevent the diagnosis of true chronic mal-behavior, they say.

Replicating results

In their Nature Methods article, the researchers call for similar scrutiny of other behavioral animal models. One important criterion is that findings from experiments conducted using an animal model must be replicable in other labs. The lack of replicability is a problem in the field, they note, which is currently insufficiently addressed by complicated and costly efforts to standardize experimental protocols.

Not every experiment needs to be replicated in many labs, sacrificing more animals, time and money, says Prof. Benjamini. "Developing a collaborative database that draws on the different experiments conducted across the globe, as well as using appropriate data mining tools, can yield the needed yardstick to check for replicability of findings for scientists in isolated labs," he adds. Such a community-based effort can help researchers better identify valid experimental results.

The Israeli Science Foundation and a European Research Council Grant supported the research.
